The PPIs reduce drug absorption and block metabolic enzymes.

Certain combinations may lead to –

  • Reduced efficacy: Esomeprazole reduces drug absorption. Avoid its use with Clopidogrel, Antifungals, Antivirals, and Heart failure medications.
  • Increased toxicity: Esomeprazole reduces drug clearance; it slows down drug metabolism. Avoid with Anti-seizure medicines, Immunosuppressants, and Warfarin.
  • Cardiac toxicity: Domperidone leads to QT prolongation. It must be strictly avoided with cardio-toxic drugs such as HIV medicines and Antipsychotics.
  • GIT side effects: Increased risk with anticholinergic and opioid drugs.
