It is a drug that is consumed orally. It’s covered by an external coating that gets easily dissolved when it enters the intestines. Each of the tablets is designed to hold 150 mg of Lamivudine. One of the main uses of this drug is for anti-retroviral therapy for curing HIV infections. It can help to prevent the replication of HIV in the body cells.

It mainly acts as a reverse transcriptase inhibitor for the virus. The introduction of the drug in the form of monophosphate in the DNA chain of the HIV virus results in anti-viral activity, resulting in the inhibition of the replication.

Administration of Lamipure 150 (Lamivudine)

ADULTS:

When the drug is administered orally, it can be absorbed with a recorded bioavailability of nearly 80%. The mean to the maximum concentration of serum is observed to be about an hour. The therapeutic dosage levels of 4mg/kg/day in the form of two 12-hourly doses, and Cmaxis is in the range of 1-1.5 mg/ml. The mean volume of distribution has been reported in the range of 1.3l/kg. The clearance of Lamipure has been noted as 0.32 L/kg/h. The renal clearance has been noted with more than nearly 70% by tubular secretion.

The mean half time for elimination has been noted for a maximum of 7 hours. The half-life of lamivudine triphosphate is very long, with an average of nearly ten hours in the blood lymphocytes. There has been a delay in Tmax, and a decrease in Cmax has been noted when administered with food. There is no dose adjustment needed. Lamivudine is expected to penetrate the central nervous system and is also expected to reach the cerebrospinal fluid. The right extent of relation with the clinical efficacy is not known.

CHILDREN:

Generally, the effect of Lamivudine is similar to that of adults as well. But here, a slight difference in the bioavailability is observed. There has been a reduction in bioavailability to 65%, and there is a slightly increased clearance recorded as 0.5L/kg/hr. There has been limited data availability for the paederastic patients who are below three months of age. However, in general, Lamipure shows results for the treatment of HIV infection in both adults and children. The contraindications have been regarded as sensitivity to any of the ingredients in the drug.

Safety Precautions

The interaction of the drug with Trimethoprim leads to an increase in the plasma concentration when given at the therapeutic dosage levels. So any dose adjustment is not required unless the patient is suffering from any renal impairment.

However, when Lamivudine is closely administered with Co-Trimoxazole, patients suffering from renal impairment have to be carefully noted.

2. Blood disorders like thrombocytopenia and anemia have also been reported

4. Fatigue and fever have also been reported
5. Chances of muscle system disorders
6. Pancreatitis
7. Hypersensitivity reactions like skin rashes can occur
8. A rise in serum amylase per the laboratory reports
