Tudobest-L (Tenofovir Disoproxil Fumarate 300mg & Lamivudine 300mg Tablets)

This drug supports anti-viral activity and is expected to show positive results for HIV-1 infected people. It is soluble in water. The bioavailability was recorded for about 25%. The in-vitro binding of the drug to the plasma of the human body was observed to be less than 0.7 and 7.2%. The steady-state distribution was recorded at 1.3 with a tolerance of 0.6L/kg. Around 70% of the dose was observed to be recovered in the urine as an unchanged component within 72 hours of dosing. In the single dosing, the drug’s half-life was observed to be 17 hours during the oral administration of the drug.

Administration of Tudobest-L (Tenofovir Disoproxil Fumarate 300mg & Lamivudine 300mg Tablets)

Lamivudine is a drug that selectively inhibits the activity of HIV-1 and HIV-2. The metabolization of Tudobest-L (Tenofovir Disoproxil Fumarate 300mg & Lamivudine 300mg Tablets) occurs intracellularly to the active 5-triphosphate that helps in the inhibition of the RNA and the DNA-based activities of the HIV reverse transcriptase, which is performed by the termination of the DNA viral chain. Lamivudine does not interact with the metabolism of deoxynucleotides and hence hardly develops an effect on the mammalian cells and the mitochondrial DNA. In the case of adults, Tudobest-L (Tenofovir Disoproxil Fumarate 300mg & Lamivudine 300mg Tablets) is well-absorbed, with a bioavailability of nearly 80% observed. The mean to maximum concentrations of serum has been noted for up to an hour. The half-life for the elimination concentration of serum is observed in the range of five to seven hours. The mean systemic clearance was 0.32 L/kg/h, with a renal clearance of more than 70% obtained via tubular secretion. Lamivudine shows a limited binding in the case of albumin and shows a linear peak in plasma levels. Dose adjustments aren’t required since it does not add any further significance; the patient’s safety is considered.

Drug Dosages

In the case of adults more than 12 years of age, one tablet a day is recommended. In children ranging from 3 months to 12 years of age, 4 mg/kg twice a day is recommended. A maximum of 300 mg is the final limit to the dosage that could be taken. However, no specific dosage recommendations have been proposed for children below three months, and there is a limitation on the data available in this regard. The drug can be consumed along with or even without food. There is no specific condition on consuming it either way.

Drug Precautions

The usage must be very careful about patients suffering from advanced cirrhotic liver disease due to chronic Hepatitis B infection. There are chances of rebound after the treatment of hepatitis. If, in any case, drug overdosing occurs, special monitoring of the patient has to be done to check for toxicity, and supportive treatment has to be carried out as necessary. The shelf life of the drug is about 24 months from the manufacturing date of the drug.

Side effects of Tudobest-L (Tenofovir Disoproxil Fumarate 300mg & Lamivudine 300mg Tablets)

1. Pancreatitis has been noted in patients who received Tudobest-L (Tenofovir Disoproxil Fumarate 300mg & Lamivudine 300mg Tablets).
2. The long-term use of Tudobest-L (Tenofovir Disoproxil Fumarate 300mg & Lamivudine 300mg Tablets) can result in lactic acidosis.
3. Sudden ending of the therapy is recommended in patients with an increased level of lactate. Therefore, treatment with lamivudine should be stopped if the symptoms are indicative of lactic acidosis or hepatoxicity.
4. Antiretroviral therapy has not been able to prove that it could prevent the transmission risk of HIV to others, so accurate precautions should be continuously encouraged.
5. In patients with moderate to severe renal impairment, there are chances of an increase in the half-life of lamivudine, so the dosages should be carefully monitored and adjusted.
